Asp f3, located in peroxisomes, belongs to the highly conserved family of peroxiredoxins involved in redox homeostasis and response to oxidative stress. Asp f3 is a major allergenic component of the environmental mold Aspergillus fumigatus, a causative agent of allergic fungal diseases affecting the upper and lower respiratory tracts, such as allergic fungal rhinosinusitis, severe asthma with fungal sensitization, and allergic bronchopulmonary aspergillosis. Asp f3 shows cross-reactivity with homologs from other species and genera of fungi.
Asp f3 is a 19 kDa protein that can form dimers through two disulfide bonds. It participates in cellular redox homeostasis and response to oxidative stress. Asp f3 also plays additional roles in the fungal cell, such as maintaining cell growth under iron starvation conditions. Due to its ability to detoxify reactive oxygen species and prevent growth arrest under iron limitation, Asp f3 is a virulence factor of A. fumigatus and a vaccine candidate. It has confirmed allergenic activity and sequence identity of 80% or more with homologs from the Aspergillus, Candida, and Penicillium families.
