Protamine (protamine sulfate) is a coagulant, hemostatic agent, and heparin antidote. It exhibits a specific anti-hemorrhagic effect in bleeding caused by heparin overdose. The test determines the level of sensitization to protamine, aiding in the diagnosis of allergic reactions.

The severity of reactions ranges from mild to life-threatening. Risk factors include insulin-dependent diabetes mellitus, vasectomy, fish allergy, prior exposure to protamine, and infusion rate.
